Cell-Free Protein Synthesis (CFPS)

CFPS is a synthetic biological technology. The technology uses the molecular translation mechanism in cell extracts to complete protein expression within a day. CFPS is an ideal method for functional protein translation in vitro. The cell-free gene expression reaction system consists of three parts:

  • Cell Extract —— Contains ribosomes, RNA polymerase, and other transcription and translation accessory proteins derived from a source strain.
  • Reaction Mixture —— Contains supplemented cofactors for protein synthesis (like amino acids, nucleotides, salts, polyamines, an energy source, molecular crowding agents, metabolic cofactors, buffer, transfer RNAs, and other metabolic additives)
  • DNA Template —— Plasmid DNA or linear expression template obtained from PCR

The standard reaction volume is on the order of 10μL. The typical protein yield in the reaction is hundreds of mg/ml.

Achievable Goals Based on the Platform

  • Using the Cell-Free Protein Synthesis Technology, we can achieve the following goals for our clients:
  • Expression conditions optimization. The lack of cell wall provides an open and universal environment for active monitoring, rapid sampling and direct manipulation of protein synthesis, which can artificially control the reaction conditions and process.
  • Greatly shorten the expression period. The cell-based expression process takes at least 2-3 days in general, while cell-free expression requires only 1-2 hours.
  • It can express proteins that are difficult to express, such as membrane proteins and toxin proteins.
  • Preparation of protein complexes protein-protein interaction studies.
  • Insertion of unnatural amino acids at specific sites, which is of great significance to the research and development of recombinant protein drugs.
