This BmNPV system has the same working mechanism as AcMNPV while it shows a greater capacity of rapid recombinant protein expression. BmNPV Bac-to-Bac system requires neither virus amplification, nor preparation of recombinant bacmid solution for transfection. Thus, this system can dramatically shorten the expression period.
A BmBacmid is assembled with reference to AcMNPV Bacmid. It harbors a large fragment that contains mini-F replicon, Kanamycin resistance marker, lacZαpeptide segment and a bacterial transposon targeting sit (mini-attTn7). Through BmBacmid, the expression vector construction can go efficiently.

Polyhedrin locus is knocked out, resulting in the failure of virus inclusion body formation. BmBacmid is highly infectious for hemocoel tissues and cultured cells, requiring neither transfection nor virus amplification step.
The expression procedure
Donor vector subclone construction, vector transformed to E. coli cells, extraction of the recombinant BmBacmid, injection of the BmBacmid to silkworm larvae, harvest of target protein.
Application
Suitable for small-scale fast protein production. The recombinant protein can be harvested within 4 weeks.
Polyhedrin remained to ensure the formation of inclusion body which makes virus or the BmBacmid highly infectious to the midgut epithelial cells and mediate infectious to larva transmission. Silkworm can be easily infected by eating mulberry leaves sprayed with recombinant BmBacmid.
The expression procedure
Donor vector subclone construction, vector transformed to E. coli cells, extraction of the recombinant BmBacmid, BmBacmid transfect silkworm cell to form virus particles, BmBacmid extraction and feed to silkworms, harvest of target protein.
Application
It saves the time of injecting silkworms one by one, thus is suitable for large-scale protein production.
