Biolistic Bombardment Nucleic Acid Delivery System

Biolistic bombardment has been widely used in DNA transfer in the study of transient gene expression to understand gene expression and function in plants. However, the transfection efficiency of traditional biolistic bombardment is relatively low, and multiple copies will be occasionally integrated. This fact makes the genetic modification of plants more complicated.

To address the problem, Creative BioMart has developed an enhanced biolistic method for high-throughput single gene insertions process.

Technology Description

We use polyethylene glycol (PEG) and magnesium salt solution instead of spermidine and calcium chloride in the standard coating method to precipitate the target sequence onto gold particles. The particles are used for the transgenic culture of plant callus. Once the DNA is introduced into the target cell, although it may not be integrated into the host genome, it can still be temporarily expressed until it is degraded by the cellular nuclease. The frequency of transient expression is higher than that of stable transformation and does not depend on the whole plant regeneration. The transient expression system obtained by this method has a more stable conversion rate.

The nucleic acids used for bombardment can be DNA (usually convenient and stable circular plasmids) controlled by the eukaryotic promoter and terminator sequences compatible with plants, or linear DNA or PCR products, or RNA

Through this method, the target sequence can be delivered directly to various tissues, such as embryogenic callus, immature embryo and pollen grains.
