Saccharomyces cerevisiae has become an important tool for protein expression research, industry and medical applications. As a food organism, it is useful for protein production. The yeast can grow rapidly on a simple medium, with high cell density, and its genetics is more advanced than other eukaryotes, so yeast can be manipulated almost as easily as E.coli. As eukaryotic, Saccharomyces cerevisiae is a host organism suitable for high-level production of secreted and soluble cytoplasmic proteins.

In our technology platform, using Saccharomyces cerevisiae to express protein involves the following steps:

  • Using competent E. coli cells to absorb the target DNA sequence
  • Integrate DNA into bacterial genome or cyclize DNA sequence to exist as plasmid
  • Selection of transformed E.coli using selective markers (antibiotics)
  • The selected Escherichia coli is amplified in a suitable medium
  • Isolation of DNA or plasmid
  • Transformed into yeast
  • Screening transformants to integrate DNA into yeast chromosomes
  • Selection and amplification of high expression yeast clones in a suitable medium
  • Isolation and purification of intracellular/secretory proteins

Saccharomyces Cerevisiae Protein Expression Service

Based on the basic expression principle above, we offer four types of expression vectors for customers to choose from. Each vector has its features and applications.

Yeast Integrating plasmids (YIp)——The plasmid lacks ori, thus must be directly integrated into the host chromosome by homologous recombination.

Yeast Centromere plasmids (YCp)——It is considered as a low copy vector and combines partial ARS and partial centromeric sequence (CEN)

Yeast Episomal plasmids (YEp)—— It is considered to be a high copy vector which is similar to bacterial plasmids. A fragment from the 2-micron circle allows for 50+ copies to stably propagate per cell.
