Lac E.coli expression system is designed and constructed based on Lac operon regulatory mechanism of E.coli. The Lac operon is one of the earliest applied operons for the expression of foreign proteins, and now the Lac operon regulated E. coli expression system has been widely used for experimental and industrial purposes.

Lac lactose operon is composed of promoter Plac + manipulation gene LacO + structure gene. Its transcription is positively regulated by CAP and negatively regulated by lacI. The lac operon has a polycistron structure. In the absence of a downstream transcriptional regulator, lacI is tightly bound to the promoter of the gene. In the presence of an inducer (IPTG/lactose/temperature), after binding with repressor protein, the binding ability of lac operon is reduced and dissociated, so the transcription of lac operon was activated.

IPTG induction can obtain ideal protein yield, but some countries have standardized the production pathway of recombinant protein for human use. If your target product is only for laboratory use, you may consider an IPTG-induced lac system.
IPTG is commonly used as an expression inducer in many E. coli expression systems. However, IPTG is expensive and potentially toxic, consequently neither suitable for large-scale protein expression nor medical protein production. For clients who want to avoid IPTG in the expression system, please consider the lactose-induced Lac system.
