The inner layer of the Saccharomyces cerevisiae cell wall is mainly composed of dextran skeleton, which contains GPI (glycosylphosphatidylinositol) anchored regions. By fusing the heterologous target protein gene sequence with the cell wall protein flocculin sequence and introducing it into yeast cells, the target protein can be localized and expressed on the yeast cell surface by the mechanism of protein transport to the membrane surface (GPI anchoring). In Creative BioMart's yeast expression platform, clients can find the surface display service mediated by flocculin. We offer two display solutions for clients to choose from: the GPI anchoring system and the flocculation domain anchoring system.
The GPI system includes 8 kinds of cell surface display systems with GPI anchor constructed by the C-terminal of Flolp. According to the characteristics of the target protein, the length of the flop peptide is determined, and then the C-terminal of the target protein is fused with the anchor sequence
The flocculation domain anchoring system uses the adhesion ability of Flo1p flocculation domain for surface display. The system includes Flo1p flocculation domain (FS and FL proteins) and secretion signal and insertion site of the target protein. The N-terminal of the target protein is fused with Flo1p flocculation domain, and the resulting protein is thought to induce cell adhesion through non-covalent interaction between Flo1p flocculation domain and the cell wall mannan chain.
