The basic principle of E. coli cell-free protein synthesis system is:

  • Plasmid DNA or PCR products are used to synthesize mRNA under the action of RNA polymerase
  • The translation factors, enzymes, amino acids, energy and tRNA of E. coli extract are used to translate mRNA into protein
  • mRNA is reused to synthesize protein in E. coli extract
