Technology Overview

The reaction system is a transcription/translation-coupled system that is competent for up to 0.1 mg/ml protein within 2 hours. The system is quick, convenient and simple. It consists of:

• Circular or linear DNA that contains SP6 RNA polymerase promoter is used as the template
• Optimized wheat germ extract and contains all the components (tRNA, ribosomes, amino acids, SP6 RNA polymerase, and translation initiation, elongation and termination factors) necessary for protein synthesis directly from DNA templates

The wheat germ extracts are able to provide co- and post-translational modifications such as phosphorylation, farnesylation and myristoylation. The signal sequence recognition and targeting can be achieved by adding the signal recognition particle and microsomes to the extract. If you want to conduct glycosylation, we will complete it by the addition of canine microsomal membranes to the extract.

The synthesized protein can be analyzed by SDS-PAGE or used directly in numerous applications.
