Cell-free protein synthesis system (CFPS) has become a popular method of protein synthesis. CFPS systems can be divided into two categories: cell extract-based and recombinant systems. The cell extract-based systems (including E.coli CFPS, Rabbit Reticulocyte Lysate CFPS, Wheat Grem Extract CFPS, Insect CFPS, and Mammalian CFPS) are powerful but complex and uncertain. For example, the rapid shortage of energy or degradation of nucleic acid and protein may lead to poor reproducibility of the system. The uncertainty of extract components and the lack of information about system components will affect the reliability of the system.
Protein synthesis Using Recombinant Elements (PURE) system has been developed to solve the problem. The PURE system is established based on the E.coli CFPS, however, it consists of only purified factors necessary for transcription, translation and energy regeneration.
