PURE Cell-Free Protein Synthesis Service

Cell-free protein synthesis system (CFPS) has become a popular method of protein synthesis. CFPS systems can be divided into two categories: cell extract-based and recombinant systems. The cell extract-based systems (including E.coli CFPS, Rabbit Reticulocyte Lysate CFPS, Wheat Grem Extract CFPS, Insect CFPS, and Mammalian CFPS) are powerful but complex and uncertain. For example, the rapid shortage of energy or degradation of nucleic acid and protein may lead to poor reproducibility of the system. The uncertainty of extract components and the lack of information about system components will affect the reliability of the system.

Protein synthesis Using Recombinant Elements (PURE) system has been developed to solve the problem. The PURE system is established based on the E.coli CFPS, however, it consists of only purified factors necessary for transcription, translation and energy regeneration.

Based on the originally PURE system, we have made some improvements to enhance the stability and expression efficiency.

  • The PURE mixture consists of:
  • Aminoacylation factor: amino acid, tRNA
  • Energy regeneration factor: CK, NDK, ADK
  • Transcription factors: NTPs RNA polymerase
  • Translation factors:  IF1/IF2/IF as initiation factors, 3EF-Tu/EF-Ts/EF-G as release factors, ribosome recycling factor (RRF), 20 kinds of aminoacyl-tRNA synthetase (ARS), methionyl-tRNA transformylase.

Technology Principle

The target protein can be easily expressed by simply adding the DNA template to the PURE mixture. It can be easily engineered according to purposes or the proteins to be synthesized, by manipulating the components in the system.

If you want to select our PURE system to express your target protein, here are some tips for your template design.

  • Both circular and linear DNA will work for the system as long as it contains a T7 promoter and a ribosome binding site.

  • For the circular template, the T7 terminator is required at the downstream of the stop codon.
  • For the linear template, the T7 terminator is not necessarily required at the 3'-terminus.
