Phage Surface Display Platforms

Three surface platforms based on different phage species can be found at Creative BioMart. In addition to the most frequently used filamentous phage M13, we also offer T4 and T7 phage for clients to choose to produce antibodies or protein.

M13 Phage Display Platform

M13 phage is E. coli specific filamentous phage, which infects the host by contacting with pili. It is composed of protein coat and its coated circular single-stranded DNA. The secondary coat proteins of PIII and pVII can be fused at the N-terminal to present the desired molecules, and PVI can be used for C-terminal fusion. The platform is excellent for recombinant antibody production, antibody affinity maturation and native species antibody library construction.

To further improve the display efficiency, we have developed a novel foreign gene delivery method in M13 platform. The conventional phagemid vector is modified to contain both phage and bacterial ori for replication, a phage packaging signal, a marker gene for screening and the coat protein for fusion. The manipulated vector is easy to prepare and shows a higher yield dsDNA and better transformation rates.

However, if required, the conventional display method which directly  inserts the gene of interest into the phage genome is also available.

T4 Phage Display Platform

Bacteriophage T4 is a large, tailed, double-stranded DNA (dsDNA) virus. The foreign protein can be displayed on its surface by fusing with the highly antigenic outer capsid protein (HOC) and the small outer capsid protein (SOC). The platform is suitable for monoclonal antibody production. We fuse pathogen antigens into the N or C terminal of SOC and HOC to provide an advanced platform for producing high affinity and high specificity antibodies. It can produce nano antibody, double-antibody, chimeric antibody, VHH single domain antibody and special monoclonal antibody in a short time.

T7 Phage Display Platform

T7 phage peptides are anchored to viral coat proteins through their amino ends (N-terminus).

Phage T7 is a lytic phage, which replicates and expands in the host cell, and its progeny is released through the lysis of the host cell. Unlike filamentous system, T7 displayed peptides or proteins do not need to be secreted through bacterial cell membrane, so they are not biologically restricted as M13 phage display system. The cleavage characteristics of T7 phage also shorten the time of separating one round of bio elutriation from the next.

T7 phage display system is suitable for high copy display of peptide or protein with small molecular weight and with free C-terminus.
