Transient gene transfection system (TGE) is an attractive option for rapid (within a few weeks) productions of mg to g level protein in culture. The system is based on the ability of cells to express heterologous DNA in a short time.

More and more recombinant protein expression systems have been using Drosophila melanogaster cells. Among them, Schneider's 2 is the only cell line for heterologous gene expression. Because these cells can grow in suspension and reach high concentration, it is easy to enlarge. In addition, these cells can be transfected with vectors carrying inducible promoters, so as to design reasonable biological processes for protein production.

Creative BioMart has developed a transient transfection process based on S2 cells to allow the rapid production of recombinant proteins. Technology Description

The protocol is suitable for the secretion and expression of recombinant proteins. The host cells are S2 cells. Our expression vector contains pUC starting point of DNA replication, ampicillin selected gene in bacteria, C-terminal peptide sequence of V5 epitope and 6xHis tag. Expression vector is maintained and constructed in DH5 α E. coli. The recombinant protein will be secreted out of the cell mediated by BiP (immunoglobulin heavy chain binding protein) signal.

We use suspension culture for transfection. Compared with adherent cell transfection, suspension cell transfection has other advantages: the continuous mixing of suspension cells ensures the uniform distribution of cells and sediments, the conventional passage of cells is easier, the number of cells is higher, the density and product titer can be achieved, and the method is scalable. These characteristics confirm the importance of optimizing the expression conditions of heterologous genes in Drosophila cells, because they are easy to grow in suspension, which is conducive to their application in biological processes.
