Based on phage display technology, the construction of native, semi-synthesized and fully synthesized antibody libraries from non-immunized/immunized animals or patients can be achieved. So far, Creative BioMart had handled hundreds of antibody library construction cases, and the customer satisfaction rate is as high as 99%. Naïve antibody library is a non-immunized antibody library. The Naïve Antibody Libraries are excellent for the production of native antibodies in any species. The library can be used to isolate human monoclonal antibodies and select antibodies to cell-surface/tumor-associated antigens.

Creative BioMart’s technology team is able to construct naïve Fab, scFv and VHH libraries for our clients. To construct the library, we collected peripheral blood lymphocytes (PBL) from non immunized hosts (humans, different animals), isolated the target antibody gene, and then cloned it into phage gene fragments to form phage display library with special fragment of antibody.

To construct the library, the V-gene repertoires of heavy and light chain is inserted into the M13 phage vector so that a repertoire of Fabs can be displayed on the surface of the host. The Synthesized Antibody Library is a large-size Fab, scFv and VHH library. It is excellent for high-affinity antibody generation, isolation of abs-special antibodies, antibody selection to cell-surface/tumor-associated antigens, and antibody affinity maturation.

Construction of Fab&Fc Antibody Libraries

Fab libraries can be applied for the isolation of antibodies (Abs) with specificity for haptens, foreign antigens, and self Ags. We construct the library by cloning the LC and HC variable region genes into a phagemid vector and displaying it on the phage surface.

The Fc fragment of antibody can be used to treat acute immune diseases instead of intravenous immunoglobulin. Based on the phage surface display technology, we can provide naïve, synthetic and Fc-chimeric fusion protein libraries for our clients.

Construction of scFv Antibody Libraries

In order to construct scFv antibody library, we first extracted total RNA or mRNA from hybridoma, spleen, lymphocyte and bone marrow cells, and then reverse transcribed into cDNA as a template for antibody PCR. Then we used our unique multi-step antibody library generation strategy to construct a large-scale single-chain antibody library with high library diversity.

Single domain antibodies (sdAbs), also known as nanoantibodies, represents the smallest antibody that was proven of diagnostic and therapeutic usefulness.

Here, we offer an immunized sdAbs library construction services that is suitable for the generation of high-affinity and antigen-special VHH or NAR V antibodies.

Additionally, we also offer synthetic sdAbs library construction services that developed from the CDR1 and CDR3 of either naïve VHH or VNAR.
