Pichia pastoris is a kind of methylotrophic yeast. It is one of the most effective expression systems of exogenous protein in recent years. At present, many foreign proteins have been successfully expressed in the host system.

Basic Expression Principle

Sines there is no stable additional plasmid in Pichia pastoris, the expression vector needs to be homologous recombination with the host chromosome, and the foreign gene expression framework is integrated into the chromosome to realize the expression of foreign genes. The advantage of integrated expression is to maintain the stability of foreign genes and produce multiple genes.

The classic Pichia pastoris expression vector we offer contains the regulatory sequence of alcohol oxidase gene. The main structures include:

  • 5'AOX1 promoter fragment
  • Polyclonal site (MCS)
  • Transcription termination and Polya formation gene sequence (TT)
  • Screening marker (His4)
  • 3'AOX1 gene fragment.
  • A secretory signal peptide sequence for secretory expression.

Intracellular or Secretory Expression System

Intracellular expression system

If your protein is cytosolic and non-glycosylated, you can choose to express the protein intracellularly. We offer three corresponding vectors (1) pAO815; (2) pPIC3K (3) pPUC3.5K, as well as two host strains (1) KM71; (2) GS115 for clients to choose from.

GS115 Strain: AOX1 and AOX2 genes express AOX protein (ethanol oxidase) normally, and the utilization rate of methanol is high, which is close to the wild type and belongs to the Mut+
KM71 Strain: AOX1 gene is mostly deleted, and only AOX2 could be used to encode AOX. The utilization rate of methanol becomes slower, which shows that it is Muts

Secretory expression system

Pichia pastoris has very low levels of secretory native proteins. Therefore, the secreted heterogeneous protein comprises the vast majority of the total protein in the Pichia growth medium. For the secretory expression, we offer two corresponding vectors (1) pPICZα,A,B,C; (2) pPIC9K, as well as two host strains (1) KM71; (2) GS115 for clients to choose from.

In addition to the signal peptide from the expression vector, the following signal peptide can also be selected.
