The retroviral expression system is a new highly efficient expression system of recombinant protein which can mediate the stable and long-term expression of foreign genes. It has been widely used in foreign gene expression, gene silencing, gene therapy, and biopharmaceuticals because of its simple genome structure, easy gene manipulation, high transfection efficiency, wide host range and easy separation.
The expression system is composed of the retroviral expression vector, auxiliary vector (expressing protein needed for viral packaging) and packaging cell line.
Creative BioMart's retroviral expression system mainly uses the moloni mouse leukemia virus (MMLV). The plasmid backbone has a variety of optional screening markers for customers to choose from. Plat-E cells are used as packaging cells so that the plasmid carrying exogenous gene can be packaged by simply being transferred into the packing cell, which is more convenient to operate.
A. Construction of retroviral vector: the target gene fragment is connected to the retroviral vector.
B. Retroviral packaging: the recombinant virus plasmid is directly transfected into Plat-E cells for viral packaging. The cell supernatant rich in retrovirus particles is collected. The titer of retrovirus should at least reach 1 × 106 ~ 1 × 107 Tu/ml.
C. Establishment of stable cell lines: the experimental group and the control group are used to infect the cells with retrovirus, and the stable cell lines are preliminarily screened by antibiotics, and the monoclonal cell lines are obtained by cell sorting.
