We have developed the circular plasmids pKD1 as the expression vector for the episomal expression protocol.
pKD1 is derived from K. drosophilarium and transferred into Kluyveromyces lactis. It is extremely stable in host cells with copy numbers of 70-100 / cell. pKD1 is very similar to the 2μm circular plasmid of Saccharomyces cerevisiae, and has been commercialized as a vector.
To express the target protein, the GOI will be inserted into the MCS on pKD1. After replicating in the E.coli, the recombinant expression vector will be linearized with an appropriate restriction enzyme and then transformed into the K. lactis. A strong LAC4 promoter on the vector can be used to initiate gene expression. Using the inducible promoter LAC4, we can express secretory or non-secretory proteins for our customers.
