The technology principle is very simple. The whole process includes two steps. Firstly, the template mRNA is transcribed by phage polymerase and then translated into rabbit reticulocyte lysate. Since transcription and translation reactions are separate, each reaction can be optimized to ensure that both of them reach their full potential.

Standard RRL Protein Synthesis Service

Rabbit Reticulocyte Lysate System for mRNA

For the standard reaction system, mRNA is used as a template for translation. After 1.5 hours of incubation at 30°C, the yield can reach 1–4 µg/ml.

The rabbit reticulocyte lysate we are using is prepared using a standard protocol. The lysate contains cellular components (tRNA, ribosome, amino acids, initiation, elongation and termination factors) required for protein synthesis. After acid enzyme treatment to eliminate the endogenous mRNA in the lysate, we also optimize the lysate (such as the addition of heme and calf liver tRNA). Some applications of the system include:

  • Translation from mRNA, in vitro transcripts or total RNA into proteins in vitro
  • Characterization of mRNA products
  • The secreted proteins are co-translated by adding microsomal membrane to the translation reaction

Enhanced RRL Protein Synthesis Starting from Low-expressing mRNA

Rabbit Reticulocyte Lysate System for mRNA

This is an enhanced RRL system that can provide optimization translation for low-expressing mRNA. This technique can be used to identify mRNA types and characterize their products. Compared with the standard system, it provides more flexibility of reaction conditions. The content of optimization mainly includes (1) Optimize the concentration of Mg2+ and K+ (2) Heme, addition of calf liver tRNAs; (3) Addition of Micrococcus nuclease to eliminate endogenous mRNA.

The system can modify proteins by phosphorylation, acetylation and isoprene. Some applications of the system include:

  • Optimize the translation of low expression messages.
  • Regulation of the oxidation environment and help maintain the functional activity related to disulfide bond
  • Luciferase activity can be used to detect inhibitory pollutants in RNA preparations.
