Different from traditional methods, de novo sequencing does not need to rely on any database to analyze the results. The peptide sequence can be directly deduced from the experimental MS/MS spectrum by some calculation methods. It can be used to assay the non-sequenced biological proteins, antibodies, post-translational modified (PTM) peptides and endogenous peptides.

Technology Applications

  • Sequence the peptides that do not exist in the protein library. It can be used to detect biological proteins and unknown proteins that are not sequenced, and to reverse engineer enzymes and other protein molecules.
  • It can be used to identify post-translational modifications and mutations, and accurately determine the protein sequence.
  • In combination with matrix-assisted laser desorption/ionization (MALDI) peptide fragmentation technology, protein identification can be performed on gel spot samples stained with gold or silver.
  • Combined with ESI peptide fragmentation technology, it can be used to identify complex samples such as adhesive strip samples (i.e. SDS-PAGE samples).
