Chloroplast Expression System

Plant chloroplast genetic transformation and expression system is a new recombinant protein expression system developed in recent years. Compared with traditional nuclear transformation, chloroplast genetic engineering has many advantages, such as high expression of foreign proteins, simultaneous expression of multiple genes, no gene silencing phenomenon and good environmental safety. Why Express Your Recombinant Protein in Chloroplast

Compared with the plant nuclear expression platform, tthe chloroplast expression system has several advantages:

  • The number of copies of the chloroplast genome is large, and the expression efficiency of exogenous genes is high
  • The maternal inheritance, the exogenous gene will not escape with the pollen transmission, and the biological environment is safe
  • CThe chloroplast genome contains many cis/trans structures, and multiple genes can be expressed together
  • The chloroplast is wrapped by a double-layer membrane and has no protein outflow, which provides a relatively safe biological environment for the expression of recombinant protein.

Technology Description

Based on the conventional expression protocol, we use the chloroplast specific promoter, terminator, 5 '- UTR and 3' - UTR as the regulatory sequences of foreign genes to construct the expression vector together with the foreign genes, the expression cassette of selective marker genes and the homologous recombination fragments from chloroplast. After the expression vector is introduced into chloroplast by particle bombardment or PEG method, the target gene fragment is inserted into the chloroplast genome by homologous recombination and site-specific integration. Finally, homogenous plants are obtained by selection pressure and multiple rounds of screening to achieve the integration and high-efficiency expression of foreign genes.

The transformation process is mainly divided into four steps

  • Construction of expression vector containing foreign gene
  • Introduce the expression vector into chloroplast and integrate it into its genome
  • Selection of transgenic plants that have eached the level of homogenization
  • Expression of exogenous genes in chloroplasts of transformed plants

We usually use the promoter of psbA gene and Prrn of 16S rRNA operon.
