It is the most effective way for lactic acid bacteria to deliver foreign protein as a carrier to express foreign protein and anchor it to the cell surface. However, the active recombinant Lactobacillus still has potential safety problems. A new surface display technology of lactic acid bacteria based on non-active and non-genetic modification of bacteria-like particles (BLPs) has solved this problem. BLPs is a kind of hollow particles which can remove protein, nucleic acid, lipoteichoic acid and other intracellular and extracellular macromolecules from food-grade lactic acid bacteria by heat acid treatment, leaving only the skeleton of cell wall peptidoglycan (PGN). Its size and shape are similar to living bacteria, so it is called bacteria-like particles. The foreign antigen protein is firmly bound to the surface of the peptidoglycan skeleton by fusing protein anchor (PA). The system is simple and convenient to operate. After fusion expression of PA and antigen protein in vitro, it can be incubated in Lactobacillus peptidoglycan particles pretreated by hot acid. With the help of PA, antigen display particles can be formed.
After treatment in hot acid, degradation products and acid are removed by washing with phosphate buffered saline (PBS). There are two formats of BLPs:
Format 1: made by BLPs admixed with antigens
Format 2: antigens are bound to the surface of the BLPs.
For this latter format, it is a requirement that the subunit antigens are produced as a fusion protein with the Protan tag in a suitable production organism. Mixing of an antigen-Protan solution with BLPs results in an instant, strong, and stable non-covalent binding such that BLPs are completely covered.

BLPs are prepared from non-pathogenic food-grade lactic acid bacteria. Secondly, BLPs of lactic acid bacteria are inactive, non-genetically modified organisms, and have no biological hazards.
BLPs can display foreign antibody protein with high density, and its loading capacity is far higher than that of its maternal parent
the size of BLPs is about 1 μm, which is just the ideal size for M cells on the mucosal surface to ingest foreign antigens
