Mammalian cells can achieve satisfactory post-translational modification while insect cells can produce virus particles containing target genes efficiently. BacMam is an improved approach which inherited the advantages of both mammalian and insect expression systems. The virus particles carrying the gene of interest are formed efficiently through insect cell expression approaches.
One pathway to generate the recombinant particles is the so-called "Bac-to-Bac". The workflow of Bac-to-Bac BacMam is displayed below. Bac-to-Bac is a conventional method in an insect cell system. E. coli DH10Bac cell line, insect cell line and mammalian cell line are involved in the process. The virus which contains the target gene can be generated by the principle of Bac-to-Bac expression systems. Tn7 mediated site-specific transposition is used to integrate the expression cassette from the donor vector into the baculovirus backbone (bacmid) in E. coli DH10Bac. The recombinant bacmid is then extracted via the standard procedure and transformed into the insect cell to assemble and amplify. Finally, the amplified virus stock is transduced to mammalian cell lines.
In order to be identified by mammalian cells, an hCMV-IE1 promoter (or other mammalian promoters) is previously inserted into the donor vectors and it will be integrated into the baculovirus together with other elements as well as the gene of interest.

The efficiency of BacMam transduction in a cell line can be confirmed by expressing green fluorescent protein (GFP) using BacMam. It can be used to optimize the transduction conditions (such as medium, temperature, time, etc.)
