Technology Description

Gateway is based on the integration of phage DNA into the host bacterial genome. This technology can facilitate the insertion of the gene of interest into the baculovirus genome in vitro through the LR recombination, eliminating the section of transforming bacteria, bacmid extraction, or co-transfection. Through the Gateway recombination reaction, the recombinant baculovirus genome needed for transfection can be obtained within one hour, dramatically shortening the protein expression period.

Technology Features

The particularity of this system is mainly shown in its vector—— BacMam pCMV-DEST Vector. The technology adopted both Tn7 transposition and LR recombination for rapid subclone construction.

  • Mini-Tn7 elements support the site-specific transposition between bacmid and vector
  • A CMV promoter from Cytomegalovirus is integrated into the BacMam vector to facilitate the gene expression in mammalian cells
  • Chloramphenicol resistance gene, Ampicillin resistance gene and Gentamicin resistance gene for vector selection
  • VSV-G on the Gateway vector to enhance mammalian cell transduction efficiency

In order to be identified by mammalian cells, an hCMV-IE1 promoter (or other mammalian promoters) is previously inserted into the donor vectors and it will be integrated into the baculovirus together with other elements as well as the gene of interest.
